Highaffinity oligonucleotides

Highly specific qPCR probes contain LNA-blocked conformational modifications that increase PCR performance especially in AT-rich sequences and point mutation analysis. LNA modifications form a fixed conformation of the sugar fragment formed by a methylene bridge between the oxygen atom in the 2'-position and the carbon in the 4'-position of the sugar. The fixed ribose conformation improves the efficiency of hybridization to the target, even if there are only 3-5 LNA modifications in the oligonucleotide. PCR probes can contain up to 6 LNA nucleotides to improve duplex stability with the target DNA (see modification section for more information). Highly specific qPCR probes may be the best choice for single nucleotide polymorphism (SNP) detection or other complex applications requiring increased selectivity.
