Dual-luciferase reporter assay

The creation of reporter constructs with luciferase is a method that allows to study the transcription and regulation of genes by measuring the luminescence of luciferase, which is expressed under a given promoter. Cells are transfected by a genetic construct(s), one of which contains the reporter gene under study, which is under the control of a certain promoter, and the second vector carries a control reporter gene under the control of a constitutive promoter. Analysis with double luciferase is carried out through sequential measurement of the activity of firefly luciferase and Renilla luciferase in the same sample on a luminometer. The result of this analysis is the relationship between the activities of firefly luciferase (Fluc) and Renilla luciferase (Rluc).
