GentaFAST Plasmid Microprep Plasmid DNA Isolation Kit
The kit is designed to isolate plasmid DNA from bacteria.
The protocol is based on a modified method of alkaline lysis of a bacterial culture without preliminary sedimentation of bacteria, followed by selective DNA binding from a clarified lysate on a siliconized column membrane. The technology allows accelerating the isolation process due to rapid lysis directly from the biomass with RNase A treatment of the lysate and further centrifugation.

The protocol is based on a modified method of alkaline lysis of a bacterial culture without preliminary sedimentation of bacteria. The technology allows accelerating the isolation process due to rapid lysis directly from biomass with RNase A treatment during lysate clarification and further centrifugation. At the stage of lysis, the cell walls of bacteria are destroyed, proteins and genomic DNA are denatured. Neutralization of the lysate leads to the formation of a light curd suspension consisting of the remnants of bacterial cell walls, proteins and genomic DNA, while plasmid DNA remains in solution. After application to the column membrane, sorption of plasmid DNA occurs, impurities in the form of salts, metabolites, lipids, etc. are effectively removed at the washing stage. Due to the small diameter of the column filter, contaminants are effectively removed. At the final stage, plasmid DNA is eluted from the membrane into solution. If necessary, it is possible to concentrate the sample. The protocol allows you to quickly obtain a small amount of plasmid DNA suitable for most molecular biological applications.
